Abstract
Background: Non-invasive cosmetic procedures are increasingly popular, leading to a growing demand for topical anesthetic preparations to reduce pain and improve user comfort. Lidocaine is an effective and safe local anesthetic; however, some commercial products still show a relatively slow onset of action. Objectives: To develop a formulation, establish a preparation process, and propose in-house quality standards for a topical lidocaine cream for cosmetic applications. Methods: The cream was prepared using the melt-emulsification method to obtain an oil-in-water (O/W) emulsion. The oil phase ratio, type and concentration of emulsifiers, and supporting agents were optimized based on evaluation criteria including appearance, pH, phase stability, homogeneity, and spreadability. Lidocaine content in the formulation was determined using a UV-Vis spectrophotometric method. Results: The optimal formulation consisted of lidocaine hydrochloride 7%, palm oil 12.5%, Olivem 1000 2.5%, cetyl alcohol 5%, lanolin 5%, Emarol H40 10%, glycerin 5%, and propylene glycol 10%. The UV-Vis method at 263 nm demonstrated good linearity (R² = 0.9979), precision (RSD = 1.35%), and accuracy (99.59%). The final cream met in-house quality standards, exhibited in vitro lidocaine release of 96.8 µg/mL after 90 minutes, and remained stable for three months at room temperature. Conclusion: A stable 7% lidocaine anesthetic cream was successfully developed, showing potential applications in dermatology and cosmetic procedures.
