Abstract

Background: Allium ascalonicum L., is a rich source of quercetin, which exhibits important biological activities such as antioxidant and anti-inflammatory effects, etc. However, the recovery efficiency and practical application depend on extraction conditions and appropriate dosage forms. Objectives: To investigate factors affecting the extraction of quercetin from shallot and develop capsule formulations containing the extract. Materials and methods: One-factor-at-a-time (OFAT) evaluation of ethanol concentration (50 - 90%), drug-to-solvent ratio (1:1 - 1:20 g/mL), time (14 - 22 hours), and temperature (room temperature to 70°C). Quercetin content was determined by HPLC-UV/Vis. The obtained thick extract was used to develop hard capsule formulations by optimizing the ratios of adsorbent and glidant excipients. Results: The optimal extraction conditions were 70% ethanol, a material-to-solvent ratio of 1:5 (g/mL), extraction time of 20 hours at room temperature, yielding a quercetin content of 7.31 ± 0.03 mg/g of dry extract. The capsule formulation consisted of with dried extract : Florite® = 2.25 : 1 and 3% talc produced granules with good flowability: weight uniformity, a disintegration time of 10.7 ± 1.2 minutes, and a quercetin content of 1.24 ± 0.02 mg/capsule. Conclusion: This study successfully established an optimized extraction process and developed capsule formulations containing quercetin from Allium ascalonicum L., thereby supporting future product development and commercialization.
